The former England international has undergone scans on the injury and been to see a specialist, leaving head coach Brian Barry-Murphy having to mix and match in his back line for Saturday’s crucial home clash with Bolton Wanderers.
The Bluebirds’ form has dipped of late with only two wins coming in their last eight outings.
That has allowed Lincoln City to sprint 12 points clear at the top in the race for the League One title.
But Cardiff still hold a seven-point advantage over third placed Bradford City in the battle for the second automatic promotion slot – and eight over fourth placed Bolton – with a game in hand.
As well as Chambers, Barry-Murphy also has injury concerns over Wales central defender Dylan Lawlor, while Gabriel Osho is suspended.
“Calum has had the scan and he's seeing the specialist today. We should know more on that in the next 24 hours, but he obviously couldn't continue and he wasn't even close to carrying on at half-time on Monday,” said Barry-Murphy.
“It just depends on the results of his scan and what the specialist says. So once that becomes clear I think it'll just be a pretty easy decision as to whether he can play or he can’t.

“We haven't trained since Peterborough so we'll have a look at Dylan today (Wednesday) and tomorrow.
“He's not fit to train today so we'll have to see how he is tomorrow and then take it day by day before Saturday.
“We have options at the back. Perry Ng is very flexible and did a really good job there for us at Peterborough.
“There have been many different things we‘ve had to deal with, and this is just another one which could provide an opportunity for somebody else to step in.”
With two defeats and a draw in their last three games at home it is going to be a nervy game for the players and fans against a Bolton side that beat the Bluebirds 1-0 in October.
Bolton have lost only once in their last 15 matches.
But Barry-Murphy believes the return of top scorer Yousef Salech could provide a major lift for Cardiff as they push for promotion during the final stretch of the season.
Salech made his comeback from a long-term injury off the bench in the draw at Peterborough, and his manager made no secret of how significant that moment was for both player and club.

“I think it's brilliant for him to be back after such serious injury. We're all very pleased from him, he's clearly a player of the highest quality who can score goals,” added Barry-Murphy.
Cardiff have created chances consistently in recent weeks but have struggled to convert them, making Salech’s return particularly timely as they look to turn performances into results.
While Barry-Murphy was careful not to place too much pressure on one individual, he acknowledged the value the forward brings to the side.
“We have other players who've scored many goals both with him and in his absence, but yeah, it's great to have him back.”
The Cardiff boss was also quick to stress that Salech’s impact goes beyond just goals, highlighting his importance within the group and the boost his presence provides in a crucial phase of the campaign.

“He's been an incredible player for us and somebody we value enormously and to have him back is a great boost for everyone.”
The Irish coach expects Salech to reintegrate quickly despite his time on the sidelines, pointing to the striker’s professionalism and physical condition.
“He’ll fit back in pretty quickly, he's someone who keeps himself very fit, so even though he's come back from a long time out, he'll be in great shape, physically.”
“I guess it's just about finding his game rhythm as quickly as possible based on how much training we can get into him in such a congested period.”
Salech’s return comes at a pivotal moment for Cardiff, who remain firmly in the promotion race despite a recent dip in results.
With six games remaining, Barry-Murphy is focused on ensuring his side finish strongly—and having his leading scorer back available could prove decisive.
As Cardiff prepare for Bolton, Salech may just be the man to provide the cutting edge they have been missing and give them the best possible chance of achieving their promotion ambitions.
